The Relationship Between Free Radicals and Antioxidant Enzymes
Free radicals have long been a hot research topic in life sciences. Reactive Oxygen Species (ROS) are the most common free radicals in the human body, mainly existing in the form of oxygen free radicals.
Excess ROS causes lipid peroxidation, damages the structure and function of biofilms, leads to protein denaturation, cross-linking and enzyme inactivation. It is closely associated with myocardial shock, radiation injury, atherosclerosis, immune deficiency and many other diseases.
Excessive free radicals trigger oxidative reactions, damage human tissues and cells, and accelerate aging — this is the well-known free radical aging theory proposed by Harman.
Oxidative stress induces oxidative damage to biomolecules, produces endogenous Damage-Associated Molecular Patterns (DAMPs) and releases cytokines. These activate downstream signaling pathways such as NF-κB, JAK, STAT and MAPK, further releasing cytokines and chemokines, recruiting inflammatory cells and causing systemic chronic inflammation.

Superoxide Dismutase (SOD) is widely present in organisms. As an indispensable oxygen free radical scavenger, it plays a vital role in defending against oxidative damage.
According to bound metal ions, SOD is divided into four types: Cu/Zn-SOD, Fe-SOD, Mn-SOD and Ni-SOD. Researches prove that SOD effectively eliminates oxygen free radicals, relieves various inflammations and protects against radiation damage. Among them, Mn-SOD features the highest stability and lowest toxicity.
In the mitochondrial oxidative phosphorylation electron transport system, oxygen is the final electron acceptor. When electron transfer is incomplete, high-activity ROS is generated. ROS attacks lipids, proteins and DNA, triggering neurodegeneration, aging, cancer, pulmonary fibrosis and vascular diseases, and it is also a key inducer of inflammation.

In mammals, Mn-SOD is located in mitochondria, regulating oxygen balance and protecting mitochondria from free radical damage. Studies show Mn-SOD is closely related to inflammation and cellular senescence.
Mn-SOD gene deficiency leads to severe mitochondrial damage and even fetal death, while Mn-SOD supplementation can significantly prolong survival.
